It depends, why are you alone now? Why hasn't it worked? Have you tried too little or too much? Are you the possessive kind? Are you a player? Are you a workaholic?
I can tell you many reasons why someone winds up alone. But I don't know what's your particular circumstance.
And at 46 I really don't think you will necessarily be alone forever, but it could be time to analyze what can YOU do to change this (without blaming someone else for it). Because ultimately, you can only control what you do. You cannot control what the men/women out there will do.

I'm 31 and have never asked myself this because somewhere I will always have a friend. I just was left by my 3rd long term lover who claimed he was "The One", but I can move on.
Stay positive and seek a companion but don't pin all of your hopes on them. Just be yourself! :)

What is it that you think you're missing? No one can exist in this world and be totally alone. On the one side there's the need for emotional companionship but on the other side there is the freedom of not having to relent to or fight the demands of someone else. Someone who feels that they have a part of you and sometimes take advantage of that part and get power and control of you. To me that kind of companionship is not worth losing myself over. I prefer to be alone, content, without someone else's demands being made on me. Emotional turmoil is stressful and sometimes deadly. Again, it's not worth it. I choose to be alone and I am a solid, stable person.

take heart- b/c i work in a casino and see men in their 40's (50's, 60's) finding women and having fam's all the time. women don't have that option (which ****** me off, but another topic all together)

no i'm sure you will find someone or in the hopes of true love maybe they'll find you, dont ever give up on love, or lose hope its much to beautiful of a gift.....

Have you asked yourself what you are looking for in a companion? You need to get out and meet people and time will tell the tale.Stay active and get out there and never give up on love.
